Studies on Protein Synthesis of Ethionine Induced Cirrhotic Liver in Rats

Abstract
The purpose of this work was 1) to elucidate the correlation of histological and chemical changes, and 2) to study the activity of protein synthesis, in the liver of rats which had been fed with 0.25 DL-ethionine diet for 6-9 months.
A significant increase in liver weight could be demonstrated in severe cirrhotic and cancerous livers. Protein nitrogen of liver microsome and supernatant decreased in parallel with the extent of cirrhotic change, but the hydroxyproline content, though it showed a tendency of increase in severe cirrhotic liver, lacked an ex-act correlation with histological evicience of The accumulation of C14-labelled alpha-aminoisobutyric acid in liver was not altered in slightly cirrhotic livers, but significantly decreased as the cirrhotic change extends. On the other hand, the incorporation of L-lysine-Cia into liver and plasma protein, especially in albumin fraction, showed a decrease in slightly cirrhotic liver as well as in severe cases. The livers which manifest malignant changes, showed alittle increase in incorporation of amino acid into liver protein compared to the livers of cirrhotic change alone, where the uptake of AIB was more impaired.
